Cancer immunotherapy

Cancer immunotherapy is a therapy used to treat cancer patients that involves or uses components of the immune system. Some cancer immunotherapies consist of antibodies that bind to, and inhibit the function of, proteins expressed by cancer cells. Other cancer immunotherapies include vaccines and T cell infusions.

Multifunctional metal-organic framework-based nanoreactor for starvation/oxidation improved indoleamine 2,3-dioxygenase-blockade tumor immunotherapy

Inhibited immune response and low levels of delivery inhibit starvation cancer therapies. Here, the authors report on the co-delivery of glucose oxidase and IDO inhibitor 1-methyltryptophan using metal organic frameworks and show amplified release in response to starvation therapy along with immune modulatory effects.

Patient-reported outcomes and neurotoxicity markers in patients treated with bispecific LV20.19 CAR T cell therapy

Knight et al. assess short-term patient-reported outcomes (PROs) and markers of neurotoxicity in patients with leukaemia or lymphoma treated with LV20.19 CAR T cells. In this small trial cohort, the authors report improvements in mood following CAR T infusion and identify kynurenine pathway molecules as potential markers of mood and neurotoxicity.

Claudin18.2-specific CAR T cells in gastrointestinal cancers: phase 1 trial interim results

In an interim analysis of a phase 1 trial, CAR T cells specific for Claudin18.2, a tight junction protein isoform highly expressed on gastrointestinal tract tumors, were well-tolerated and exhibited promising clinical activity in patients with gastrointestinal cancers.

CD4⁺ T cells are required to improve the efficacy of CIK therapy in non-small cell lung cancer

Herein, Liu and colleagues show that CD4⁺ T cells in cytokine-induced killer (CIK) cells are required to enhance the clinical efficacy of CIK therapy. In addition, CD4⁺ T cells help via IL-17A production is critical to restoring the function of CD8⁺/CD3⁺CD56⁺ T cells and reversing the exhaustion of PD-1⁺TIM-3⁺ T cells in CIK therapy.

A new study reveals that neoantigen cancer vaccines synergize with anti-PD-L1 ICT in a preclinical model and provides mechanistic insights into this synergy.

PIK3CA hotspot mutation generates a shared neoantigen targetable by TCR gene therapy

PIK3CA gain-of-function mutations are among the most common alterations in human solid cancers. Through the use of stimulation-induced functional T cell receptor (TCR) sequencing (SIFT-seq), a panel of TCRs that bind a mutant PI3Kα shared neoantigen was identified, including a potential clinical candidate that engages cancer cells via a distinctive CDR3β loop.

T follicular helper and B cell crosstalk in tertiary lymphoid structures and cancer immunotherapy

Tumor-infiltrating lymphocytes (TILs) are critical in the elimination of cancer cells, a concept highlighted by recent advances in cancer immunotherapy. Significant evidence reveals that their organization in tertiary lymphoid structures together with specific subpopulation composition/balances stimulates cellular crosstalk and anti-tumor immunity in patients.
